Lectin Microarray
A sensitive and high-throughput method for glycan profiling is the lectin microarray. Glycan-binding proteins (GBPs) known as lectins can interact with glycan molecules in a specific way. The specified lectins are immobilized onto a solid phase that has been chemically (epoxy, NHS ester, for example) or biologically (streptavidin, for example) activated. A glycoprotein will attach to a particular lectin when it is incubated with the lectin chip, and the binding events observed at particular lectin sites can be used to provide pictures of potential glycan variations in the sample. Lectin microarray is a useful method for glycan profiling of a therapeutic monoclonal antibody as well as for evaluating glycan variations of purified glycoproteins.
